The experienced Rosol recovered from an uncertain start on day one of the tournament to oust Argentine Schwartzman 6-4 6-4.
Rosol lost the first two games of the match, but responded to seal victory in an hour and 20 minutes after serving eight aces in a contest in which he won 81 per cent of points on his first serve.
The unseeded Czech can expect a far bigger challenge in round two when he takes on Wawrinka, who dethroned Novak Djokovic to take his first US Open title at Flushing Meadow in his last match.
German teenager Alexander Zverev, seeded five, also moved into the second round with a 7-6 (7-3) 6-4 success against wildcard Karen Khachanov.
Ricardas Berankis was the other winner on day one, dumping out Adrian Mannarino 7-6 (9-7) 3-6 6-3.
